Quantitative profiling of actin-associated proteins in human cerebrospinal fluid. Lysis of cells leads to release of cellular components, which can be used as biomarkers for disease diagnosis. Protein profiling of cerebrospinal fluid (CSF) shows promise as a non-invasive diagnostic tool for neurological disease. However, a high level of complexity poses a challenge to effectively profile proteins in CSF. We have developed a novel experimental design, termed CSF-2, to quantitatively profile proteins in CSF by using 2-DE and Mass Spectrometry (MS). We first optimized the technique to reveal differentially expressed proteins in CSF and found that five proteins are differentially expressed in lysis buffer-treated CSF, including triosephosphate isomerase, an actin-associated protein. We then used the technique to profile actin-associated proteins in normal and neurodegenerative disease CSF. Three known actin-associated proteins were identified and their CSF concentrations significantly altered between normal and neurodegenerative diseases, including Alzheimer’s disease and amyotrophic lateral sclerosis.
